Stachyose is one of the abundant carbohydrates in plants and belongs to the members of the raffinose family of oligosaccharides. It was first found in the rhizomes of Stachys tuberifera and coexists with other related oligosaccharides in miscellaneous organs of a large number of plant species. It is a tetrasaccharide that consists of sucrose with an alpha-D-galactosyl-(1->6)-alpha-D-galactosyl group attached to the 6-position of glucose. On a weight basis, stachyose is approximately 28% less sweet than sucrose. It is primarily applied as a bulk sweetener or for its functional oligosaccharide properties. In both mice and humans, stachyose has been proven to have a selective impact on the promotion of beneficial intestinal bacteria growth and the inhibition of pathogenic bacteria. CD BioGlyco provides a Biocatalytic Approach to manufacturing stachyose by constructing in vitro enzymatic systems. We conduct the cascade reactions by utilizing the cell lysate supernatant without the addition of the expensive UDP. Stachyose is produced through the sequential transfer of galactosyl units from galactinol to sucrose, a reaction catalyzed by the enzyme stachyose synthase. Our proposed system aims to reduce production costs by recycling and regenerating UDP and inositol to facilitate the conversion of sucrose into the desired product.
To avoid the need for enzyme purification and the supplementation of additional UDP, we employ cell extracts containing the expressed enzymes and a minimal amount of UDP in the reaction medium. This approach allows for effective production and ensures maintaining the catalytic activity of the enzymes. We utilize a stepwise reaction strategy to address mismatches in enzyme characteristics. In a word, we have successfully showcased an efficient method for biosynthesizing stachyose using an in vitro multienzyme system and a stepwise cascade reaction process.
