SEC is separated according to sample hydrodynamic volume, and characterization of polysaccharides according to their structural parameters (size, molar mass, distribution, and structure). The molecular sieve is utilized to enable molecules of different sizes and numbers to complete the separation of components according to the different exclusion capabilities of each component. It is the key to a better understanding of structure-property relationships, the ability to determine the true molar mass, the structure of the macromolecules, and the composition of the copolymer detectors coupled to chromatographic systems. The rheological properties and solubility of feedstocks depend on their molar mass and branching, so good characterization along the process is important. SEC analysis is well established for the determination of oligosaccharides and medium-sized polysaccharides.

Molecular properties such as weight average molecular mass (MW), average radius of rotation, and intrinsic viscosity of soluble dextran are determined by SEC, polysaccharides are commonly dissolved in the aqueous system or analyzed directly in the extracts.
