Hydrolytic Reactions

We prepare maltooligosaccharides by hydrolytic reactions that involve the use of glycoside hydrolases, such as pullulanase and isoamylase, to hydrolyze complex carbohydrates. These enzymes break down the side linkages in starch, followed by hydrolysis using various α-amylases. This method requires precise control of reaction conditions to efficiently synthesize maltooligosaccharides with the desired number of monosaccharide units.

Transglycosylation Reactions

We prepare maltooligosaccharides by transglycosylation reactions. Transglycosylation is the process by which glycosidic linkages are formed between starting units. This reaction leads to the production of a higher molecular weight product. Transglycosylation is achieved by glycosyl hydrolases or glycosyltransferases. While glycosyl hydrolases primarily catalyze other reactions, transglycosylation is induced by altering reaction conditions or using suitable acceptors other than water.

  • During the starch liquefaction process, α-amylases, acids, or physical factors are employed to break down the starch into a more fluid state. If necessary, impurities such as proteins and fats are eliminated during this stage.
  • The second step is important which involves the saccharification of starch to form maltooligosaccharides. Saccharification processes are categorized into two types: a single enzyme process that utilizes MFAse and a dual enzyme process that combines the actions of MFAse and pullulanase. In the dual enzyme system, the addition of pullulanase to the reaction system not only enhances the reaction rate but also increases the yield of maltooligosaccharides. Pullulanase and MFAse are added together before saccharification, making both enzymes act simultaneously during the saccharification process.
  • The last step in the production of maltooligosaccharides is the purification process. That process involves removing those impurities, such as unconsumed starch, maltodextrins, and enzymes.
