What Is DB

Degree of branching refers to the number of A chains in each B chain, i.e. A: B value. There are three kinds of chains in the branched chain, A, B, and C. The A chain is bound to other chains by a glycosidic bond with a reducing end, but not bound by other chains; the B chain is bound to other chains and at the same time is bound by other chains; the C chain is bound to other chains with a reducing end. There is only one C chain in each branched chain, the other end of the C chain is a non-reducing group. A and B chains only have non-reducing groups.DB is one of the most basic structural parameters of the sugar chain. Branching-based analysis schemes may provide a viable way to resolve long and complex carbohydrate chains.

DB Analysis Services at CD BioGlyco

Branching analysis of β-glycan is a key part of understanding the composition and conformational relationships of sugar chains. At CD BioGlyco, we have a variety of methods to assist and decipher highly branched polysaccharides in research. We provide the following services to help our customers quickly and efficiently determine the percentage of branching.

  • We offer an enzymatic combined capillary electrophoresis assay service. First, fluorescently label debranched branched starch, and then determine the reducing power using the capillary electrophoresis instrument's laser-induced fluorescence detector (LIF). This method reduces the number of reaction steps in the original enzymatic assay and improves the accuracy of the results.
  • MS-assisted determination has been a powerful tool for structure analysis in recent years. We isolate the product by chemical degradation to produce oligo- or monosaccharides or even smaller fragments. Then, perform mass spectrometry/flame ionization detection (MS/FID) after multi-step derivatization of the product, which is a commonly used method for structure analysis of polysaccharides.
  • Furthermore, we also provide nuclear magnetic resonance (NMR) spectroscopy to analyze the structure of various dextrans, which quickly and accurately determine their linkages.
