The Substrate Specificity of Debranching Enzyme (DE) Treatment

The DE efficiently and exclusively cut the α-1,6-glycosidic bond, thus cutting out the entire collateral to form amylose. Due to the amylopectin content in the starch being relatively high, other amylases can not hydrolyze α -1,6-glycosidic bonds in amylopectin or hydrolysis efficiency is very low. So, the α-1,6-glycosidic bond in starch becomes a hindering factor in starch hydrolysis and needs the participation of DE. According to the difference in substrate specificity, DE is divided into amylo-1,6-glucosidase, isoamylase, and pullulanase. According to the similarities and differences in protein structure, it is a typical (β/α)8-barrel structure, belonging to the GH 13 family. DE is widely used in the starch processing industry.
